Thawing Choanos

• Tubes can be thawed at room temperature in one's hand, in a water bath at 37°C or at room temperature.
• When thawed, immediately pipette into choano media. We usually add a 1mL stock to 14mL of medium.
• Because cells may not be robust when first thawed, the cultures may look overgrown with bacteria at first. Within 2-3 days, the choanoflagellates will begin to deplete the bacteria.

MX2 will probably be very robust for a week or two when thawed, and may require frequent splitting.
